No, if you are using an iPad with IOS5 and above.
As of iOS 5, it is quite feasible to run an iPad and never actually synch with iTunes. I'm doing so with my new iPad. However, in the case of some content, you may want to employ some cloud based services such as iCloud and drop box. iCloud is useful in maintaining backups if nothing else.
Similar to James Ward4 we have been using an iPhone 4s for the last 5 months without once connecting to a computer. There are some limitations with iCloud backup, though, in terms of what is backed up, the amount, and time limitations:
