Try another USB Port on the computer. But first - quit iTunes, reboot the iPad and reboot your PC - then try again.
Reboot the iPad by holding down on the sleep and home buttons at the same time for about 10-15 seconds until the Apple Logo appears - ignore the red slider - let go of the buttons.

Does your computer see it? Does it show up in the finder/my computer? If so , then your issue is just iwth iTunes. If not then:
Are you using a hub or extender? The iPad doesn't like those anymore.
Plug it directly into your computer.
Try different USB ports. A common cause is the apple mobile device driver. Usually it installs automatically, swapping out USB ports can trigger that.
Worst case scenario, uninstall and reinstall iTunes.
Are you trying to connect via wifi? Usually it's because you are not connecting to the internet via wifi.

Additional things to try.
Try this first. Turn Off your iPad. Then turn Off (disconnect power cord) the wireless router & then back On. Now boot your iPad. Hopefully it will see the WiFi.
Change the channel on your wireless router. Instructions at http://macintoshhowto.com/advanced/how-to-get-a-good-range-on-your-wireless-netw ork.html
Another thing to try - Go into your router security settings and change from WEP to WPA with AES.
